Based on the concept of a lab-on-a-chip for genetic analysis, we developed a miniaturized platform applied to the diagnosis of genetic diseases. For the first time, the method of polymerase chain reaction (PCR) is combined with the technology of acoustic biosensors, aiming at the development of a microdevice that accommodates amplification of DNA sequences and their label-free detection by means of acoustic waves. The platform comprises two distinct modules combined into a continuous device through which the sample flows. The first part comprises resistive microheaters, incorporated to a substrate, to define three temperature zones necessary for performing PCR. A serpentine continuous flow microchannel crosses these zones, while the DNA sample flowing through 25-30 repeating cycles is efficiently amplified. After the sample exits the microPCR module, it is injected in the second module where the acoustic biosensor is located for detecting amplified DNA in real time
